miRNA Sample Preparation from Biofluids
The study of basic microRNA (miRNA) function has been performed mainly through collection of tissues by invasive methods. However, in a clinical setting, more convenient and non-invasive methods are required, such as collection of peripheral blood or other bodily fluids. For those of you currently collecting sample from patients or working on development of a clinical test, we have put together some recommendations for sample collection and preservation and for RNA extraction methods from bodily fluids.Recently, it has been demonstrated that miRNAs are present in circulating blood plasma, both free circulating and within exosomes, as well as within other biofluids such as cerebral spinal fluid, urine and even breast milk6. Circulating miRNAs in biofluids represent a potential new approach for diagnostic screening. Indeed, characteristic changes in the serum or plasma miRNA profiles of several cancers or other conditions have identified unique signatures that could be exploited as novel biomarkers in the clinic.

Degradome Sequencing Workshop
MicroRNAs (miRNAs) are endogenous small noncoding RNAs that play crucial roles in the post-transcriptional regulation of gene expression in plants and animals. They function by binding to complementary mRNA molecules (targets) and acting as negative regulators of translation. This function is part of a complex web as a single miRNA might have several target genes and a single gene may be regulated by many miRNAs. Identification of these miRNA-target pairs is crucial to understanding the biology of the miRNA regulatory mechanism. Recently, degradome sequencing, a modified 5'-rapid amplification of cDNA ends (RACE) performed with next-gen sequencing, has emerged as a comprehensive method of analyzing patterns of RNA degradation. This presentation will provide an overview of this emerging method for miRNA target identification, as well as practical information for performing degradome sequencing experiments and data analysis. Diverse case study examples will be provided to illustrate the usefulness of this latest application of next-gen sequencing technology to plant miRNA research.
