INTRODUCTION : INTRODUCTION Transdermal drug delivery system are topically administered medicamentsin the form of patches that deliver drugs for systemic effects at a predeterminedand controlled rate. Anatomy of Human skin. : Anatomy of Human skin. Patch Administration : Patch Administration Placed on the skin to deliver a specific dose of medication through the skin and into the bloodstream. Through a diffusion process, the drugenters the bloodstream directly through the skin. Basic Components Of TDDS : Basic Components Of TDDS Polymer matrix / Drug reservoir Drug Permeation enhancers Pressure sensitive adhesive (PSA) Backing laminates Release liner Other excipients like plasticizers and solvents TYPES OF TRANSDERMAL PATCHES : TYPES OF TRANSDERMAL PATCHES Single-layer Drug-in-Adhesive Multi-layer Drug-in-Adhesive Drug Reservoir-in-Adhesive Drug Matrix-in-Adhesive Desirable features for transdermal patches : Desirable features for transdermal patches Composition relatively invariant in use. System size reasonable. Defined site for application. Application technique highly reproducible. Delivery is efficient. Evaluation Of Transdermal Patches : Evaluation Of Transdermal Patches Physicochemical evaluation In vitro evaluation Thickness, Uniformity of weight, Drug content determination, Content uniformity test, Moisture content, Moisture Uptake, Flatness, Folding Endurance, Tensile Strength. Skin irritation studies, Stability studies. In vivo evaluation Animal models, Human models. Advantages of transdermal dosage form : Advantages of transdermal dosage form Avoids gastrointestinal tract difficulties during absorption caused by enzymes, drug interactions with food, etc. Avoids first pass i.e. the initial passage of a drug substance through the systemic and portal circulation. Provides the capacity for multi day therapy with a single application thereby improving patient compliance. Therapy can be quickly terminated by removal of the patch from the skin.
