The Blood Coagulation Process Blood coagulation is a process that changes circulating substances with in the blood into an insoluble gel. The process requires – The coagulation factors(proteins). Ionized calcium. Phospholipids.

Heparin, low molecular weight heparin, Heparinoids- heparan sulfate. B. Oral anticoagulants I. Coumarin derivatives: Warfarin sodium,acenocoumarol. II. Indandione derivative: Phenindione. Used in vitro Heparin. Calcium complexing agents: Sodium Citrate.

Pharmacology of anticoagulant drugs: Heparin:

Pharmacology of anticoagulant drugs: Heparin Composition:- sulfated mucopolysaccharides. Mechanism of action:- Heparin binds to endothelial cell surface. Heparin activity is dependent on antithrombinIII( plasma protease inhibitor) AntithrombinIII-protease complex formation is usually very slow. accelerated by heparin. A heparin have high molecular weight have higher affinity. A heparin have low molecular weight have lower affinity.

Toxicity Of heparin:

Long term heparin use- osteoporosis Transient thrombocytopenia :frequency-25% Severe thrombocytopenia :frequency-5% Cardiovascular : severe hypotension , intracranial hemorrhage. Toxicity Of heparin

Warfarin & Coumarin:

Warfarin & Coumarin Active agents- bishydroxycoumarin Mechanism of action- Blockage of gama-carboxylation of glutamate residues in: prothrombin, factors VII,IX,X. Carboxylation reaction is coupled with oxidative deactivation of vitamin K. Effect depends: 1.partially inhibited synthesis of vitamin K dependent clotting factors. 2.Altered degradation rates of these factors.

Toxicity Of Warfarin & Coumarin:

Toxicity Of Warfarin & Coumarin Foetal abnormal bone formation. Never administered during pregnancy. Cutaneous necrosis.
